Confocal micro-Raman Spectrometer

Make: Oxford Instruments Group
Model: WITec alpha300 Series

CAI-K Confocal micro-Raman Spectrometer

Confocal micro-Raman spectrometer is an advanced analytical instrument used to obtain highly localized chemical, structural, and molecular information from materials. It combines the principles of confocal microscopy and Raman spectroscopy to enable high-resolution, non-destructive analysis at the micro- and sub-micron scale.

The key features of the instrument include a) Confocal Optics: Enhances spatial resolution by restricting the sampling area to a small focal volume, allowing depth profiling and reduction of background signals; b) Raman Spectroscopy: Utilizes inelastic scattering of laser light to provide a molecular fingerprint of the material, revealing vibrational modes of chemical bonds and c) Micro-analysis Capability: Equipped with a microscope for precise targeting and mapping of microscopic features in samples.

The advantages of the system include High spatial resolution (typically ~1 µm lateral and ≤2 µm depth), non-destructive and label-free analysis, ability to analyze opaque, transparent, or heterogeneous materials, chemical imaging and mapping capabilities for composition variations, and suitable for solids, powders, thin films, biological samples, and polymers.

Applications

Confocal micro-Raman spectrometer offers precise, non-destructive molecular characterization with three-dimensional spatial resolution. It is an essential tool in research and industry for understanding material composition, structure, and chemical changes at microscopic scales.

    1. Materials science – crystal structure analysis,stress/strain mapping, phase identification
    2. Semiconductors – defect analysis, layer characterization, strain in devices
    3. Pharmaceuticals – polymorph differentiation, ingredientmapping in tablets
    4. Life sciences – cellular biochemical imaging without dyes or stains
    5. Geology – mineral identification, fluid inclusion analysis
    6. Forensics & cultural heritage – pigment identification, trace evidence analysis

Specifications

  • Accommodates samples up to \(120\text{\ mm}\times 120\text{\ mm}\) and \(25\text{\ mm}\) in height, with options for larger heights.
  • Research-grade optical microscope with a turret capable of holding up to 6 objectives.
  • Objective lenses include options like \(10x\) (NA 0.25), \(40x\) (NA 0.6), \(50x\) (NA 0.8), and \(100x\) (NA 0.9).
  • Ultrahigh-throughput UHTS spectrometers -Fiber-coupled ultrahigh-throughput optical instruments (up to 70 % transmission)

Instructions

Sample should be stable and not too thick, as the laser needs to pass through it or reflect from it.

Sample Requirement

Sample requirements for a WITec alpha300 series confocal micro-Raman spectrometer are flexible, but the sample must be compatible with the microscope's stage, which typically accommodates sizes up to 120 mm in x- and y-direction by 25 mm in height, with adapters available for larger heights.
